بازدارندگی تخمریزی، کاهش نتاج و وزن توسط Callosobruchus maculatus (F.) در حبوبات تیمار شده با دو فرمولاسیون نانوسیلیکا
در این پژوهش تأثیر فرمولاسیون های Aerosil® و Nanosav نانوذرات سیلیکا بر بازدارندگی تخمریزی سوسک چهار نقطه ای حبوبات،Callosobruchus maculatus (F.) در لوبیا چشم بلبلی، ماش سیاه، ماش سبز، نخود و عدس با غلظت های: 50، 100، 200 و 300 میلی گرم بر کیلوگرم بررسی شد. بذور پس از تیمار، در معرض حشرات کامل (نر و ماده) یک روزه قرار گرفتند و 5 روز بعد تعداد تخم ها شمارش گردید. بیشترین بازدارندگی در ماش سیاه با میانگین 96 درصد در غلظت 300 میلیگرم بر کیلوگرم از Aerosil® مشاهده شد. در آزمایش دوم تأثیر این فرمولاسیون ها بر تفریخ تخم و ظهور حشرات کامل بررسی گردید. بذرهای حبوبات حاوی یک عدد تخم یک روزه با غلظت های ذکر شده در بالا تیمار گردید. کمترین میزان تفریخ تخم و ظهور حشرات کامل در عدس به ترتیب با میانگین 13 و 1 درصد، در غلظت 300 میلی گرم بر کیلوگرم از Aerosil® مشاهده شد. در آزمایش سوم کارآیی ترکیبات ذکر شده بر کاهش وزن و تعداد نتاج پس از 3 ماه بررسی شد. حبوبات مورد نظر با غلظتهای بیان شده در آزمایش اول تیمار و در معرض حشرات کامل (نر و ماده) یک روزه قرار گرفتند. کمترین کاهش وزن در عدس با میانگین 10 درصد بوسیله غلظت 300 میلی گرم بر کیلوگرم Nanosav ایجاد شد. کمترین تعداد نتاج ظاهر شده نیز در عدس با میانگین 40 عدد در غلظت 300 میلی گرم بر کیلوگرم از Nanosav دیده شد. هر دو فرمولاسیون می توانند به طور مؤثر در مدیریت تلفیقی C. maculatus استفاده گردند.

تعیین زمان مناسب استفاده از قارچکشها برای کنترل بیماری لکه قهوهای جو
بیماری لکه قهوه ای (Bipolaris sorokiniana)، یکی از بیماری های برگی مخربی است که در شرایط همه گیری خسارت قابل توجهی به جو وارد می کند. استفاده از قارچکش ها، روش سریع کنترل این بیماری در سال های همهگیری می باشد. بدین منظور آزمایشی در قالب طرح کرت های خرد شده در چهار تکرار طراحی گردید. عامل اصلی آزمایش را ارقام(صحرا و یوسف) و عامل فرعی را تیمارهای سمپاشی با قارچکش های رایج منطقه شامل تیلت، فولیکور، فالکن و رکس دو به ترتیب در چهار زمان T0 (مرحله اوایل ساقه روی تا تشکیل گره 1)، T1(مرحله تشکیل گره 3-2)، T2 (ظهورکامل برگ پرچم) و T3 (گلدهی) تشکیل دادند. در تحقیق حاضر، ده تیمار سمپاشی بر اساس زمان مراحل رشدی جو در نظر گرفته شد که شامل تیمارهای Tr1: یک بار سمپاشی در مرحله اوایل ساقه روی تا تشکیل گره 1، Tr2: یک بار سمپاشی در مرحله تشکیل گره 3-2، Tr3: یک بار سمپاشی در مرحله ظهور کامل برگ پرچم، Tr4: یک بار سمپاشی در مرحله اوایل گلدهی، Tr5: دو بار سمپاشی در مرحله تشکیل گره 3-2 + ظهور کامل برگ پرچم، Tr6: دو بار سمپاشی در مرحله ظهور کامل برگ پرچم + اوایل گلدهی، Tr7: دو بار سمپاشی در مرحله اوایل ساقهروی تا تشکیل گره 1 + تشکیل گره 3-2،Tr8 : سه بار سمپاشی در مرحله تشکیل گره 3-2+ ظهور کامل برگ پرچم+ اوایل گلدهی، Tr9: چهار بار سمپاشی در مرحله اوایل ساقهروی تا تشکیل گره 1+ تشکیل گره 3-2+ ظهور کامل برگ پرچم+ اوایل گلدهی وTr10 (تیمار شاهد) بودند. در این بررسی تیمارهایی با بیش از یک بار سمپاشی ازمرحله تشکیل گره 3-2 تا گلدهی (تیمارهای Tr5، Tr6، Tr8 و Tr9) توانستند سطح زیر منحنی پیشرفت وقوع بیماری (AUDPC-I) را در رقم صحرا بین 1/86-8/76 درصد و در رقم یوسف بین 2/81-3/68 درصد کاهش دهند، در حالیکه مقدار سطح زیر منحنی پیشرفت شدت بیماری (AUDPC-S) به ترتیب در ارقام صحرا و یوسف بین 7/65- 62 درصد و 7/62-1/60 درصد کاهش یافت. بیشترین درصد افزایش عملکرد نیز در تیمارهایی با بیش از یک بار سمپاشی از مرحله تشکیل گره 3-2 تا گلدهی وجود داشت که به ترتیب در رقم های صحرا و یوسف بیش از 37 درصد (8/1092 تا 1/1284 کیلوگرم در هکتار) و 41 درصد (8/1167 تا 1290 کیلوگرم در هکتار) افزایش عملکرد مشاهده شد. بر اساس نتایج بدست آمده، تیمار Tr5 (دو بار سمپاشی در T1+T2) مناسب ترین تیمار از نظر زمان استفاده قارچکش برای کاهش وقوع و شدت بیماری، افزایش عملکرد و سود اقتصادی بود.

نماتدهای انگل گیاهی خانوادههای Merliniidae، Telotylenchidae و Pratylenchidae (Nematoda: Tylenchoidea) در گیاهان مهم زراعی منطقه ی گرگان
به منظور شناسایی نماتدهای خانوادههای Merliniidae، Telotylenchidae و Pratylenchidae در گیاهان مهم زراعی منطقهی گرگان، طی سالهای 1394-1393، تعداد 64 نمونه ی خاک و ریشه ی گیاهان مهم زراعی از مناطق مختلف منطقه ی گرگان جمعآوری گردید. پس از استخراج و انتقال نمونهها به گلیسیرین، اسلایدهای میکروسکوپی دائمی تهیه شده و اندازهگیریهای لازم انجام گردید. سپس شناسایی نماتدها با استفاده از ویژگی های ریخت شناختی و ریخت سنجی و با کمک کلیدهای شناسایی معتبر صورت گرفت. در این بررسی تعداد 11 گونه از هفت جنس شامل گونههای Amplimerlinius macrurus، Merlinius brevidens، M. microdorus، Paramerlinius hexagrammus، Tylenchorhynchus brassicae، T. crassicaudatus، Pratylenchoides ritteri، Pratylenchus neglectus، P. sefaensis، P. thornei وZygotylenchus guevarai مورد شناسایی قرار گرفت. گونهی T.crassicaudatus برای اولین بار از ایران گزارش و شرح داده میشود و اختلاف آن با گونه های نزدیک شامل T.agri، T. thermophilus، T. paratriversus،T.kashmirensis و T.mashhoodi مورد بحث قرار گرفته است.

بررسی خسارتزایی سوسک برگخوار غلات Oulema melanopus (L.) (Coleoptera: Chrysomelidae) در مزارع گندم
لاروهای سوسک برگخوار غلات Oulema melanopus با تغذیه از برگ پرچم گندم موجب خسارت می گردند. این تحقیق با هدف بررسی امکان خسارت زایی این آفت روی گندم در سه مزرعه استان مازندران واقع در نکا (ایستگاه بایع کلا)، ساری (ایستگاه دشت ناز) و جویبار (ایستگاه پهناب) در طی سال های 1392 و 1393 به اجرا در آمد. در هر مزرعه 10 نقطه تعیین و همزمان با اتمام فعالیت تغذیه ای لاروها، از هر نقطه 10 ساقه ی آفت زده و 10 ساقه ی سالم به طور تصادفی انتخاب و شماره گذاری گردیدند.از برگ پرچم ساقه های آفت زده عکس تهیه شد و با استفاده از نرم افزار UTHSCSA ImageTool for Windows Version 3 شاخص خوردگی سطح برگ پرچم در تصویر نمونه ها در 4 رده (10٪>، 10-20، 20-40 و 40٪<) گروه بندی شدند. در فصل برداشت، خوشه ساقه های علامتگذاری شده جمع آوری و تعداد دانه و وزن دانه ی موجود در هر خوشه و وزن هزار دانه اندازه گیری گردید. نتایج نشان داد که فعالیت تغذیه ای لاروها می تواند بر وزن و تعداد دانه در هر خوشه و وزن هزار دانه تاثیر منفی گذاشته و با افزایش درصد خوردگی سطح برگ پرچم، وزن دانه و تعداد دانه در خوشه کاهش یافت. در رده 40٪< خوردگی برگ پرچم، کاهش وزن دانه، تعداد دانه و وزن هزاردانه در گیاهان خسارت دیده در مقایسه با گیاهان سالم در سال 1392 به ترتیب 58/42%، 86/25% و 76/32% و در سال 1393 به ترتیب 42/32%, 59/15% و 82/19% بود. ولی از آنجائیکه شاخص خوردگی عموماً در رده های کمتر از 10٪ و 10-20٪ قرار داشته و بندرت به رده بیش از 40٪ رسید، و همچنین تعداد ساقه های با برگ پرچم آفت زده در واحد سطح پایین بود، خسارت زایی این آفت در مناطق مورد آزمایش پایین تا متوسط بود.

پاسخ بویایی سن شکارگر Orius albidipennis و زنبورهای پارازیتوئیدEretmocerus mundus و Eretmocerus eremicus نسبت به سفیدبالک پنبه Bemisia tabaci و دشمنان طبیعی رقیب
سفیدبالک پنبه(Gennadius) Bemisia tabaci یکی از آفات مهم محصولات زراعی، جالیزی و زینتی میباشد. زنبورهای پارازیتوئید Eretmocerus mundus Mercet و Rose and ZolnerowichEretmocerus eremics و همچنین سن شکارگر (Rueter)Orius albidipennis از جمله دشمنان طبیعی میباشند که به عنوان عوامل کنترل زیستی برای مهار سفیدبالک پنبه مورد استفاده قرار گرفته و نقش مهمی در کاهش استفاده از سموم شیمیایی داشتهاند. در این بررسی، آزمایشهایی توسط دستگاه بویایی سنجی برای تعیین تأثیر رایحه تولید شده از برگ خیار آلوده به سفیدبالک پنبه در جلب دشمنان طبیعی انجام گرفت. همچنین تاثیر حضور یک دشمن طبیعی در لکه میزبان، بر جلب یا دفع دشمن طبیعی دیگر مورد بررسی قرار گرفت. نتایج بدست آمده از آزمونبویاییسنجی نشان داد که برگهای خیار آلوده به پورههای سفید بالک، هر سه گونه دشمن طبیعی شامل سن O. albidipennis و زنبورهای پارازیتوئید E. mundus و E. eremics را بیشتر از برگهای سالم به سمت خود جلب نمودند. نتایج تأثیر حضور زنبور هایپارازیتوئید بر انتخاب و ترجیح سن شکارگر O. albidipennis نشان داد که این سن از برگهایی که زنبورهای پارازیتوئید روی آنها حضور دارند دوری نمود، اگرچه حضور سن شکارگر O. albidipennis روی برگها به عنوان رقیب، اختلالی در جذب این دو گونه زنبور به برگهای آلوده ایجاد نکرد.

بررسی کارآیی چهار روش استخراج DNA از زنبورعسل کوچک Apis florea (Hymenoptera: Apidae) جهت انجام آزمایشات ژنتیکی
استخراج DNA اولین مرحله در انجام آزمایشات ژنتیکی است، لذا کیفیت و کمیت مناسب DNA استخراج شده در مطالعات مولکولی امری بسیار ضروری می باشد. از این رو دستیابی به روش یا روش هایی که به سهولت استخراج DNA از زنبورعسل کوچک Apis florea Fabricius که یکی از دو گونه زنبورعسل موجود در ایران است کمک کند، می تواند حائز اهمیت باشد. این گونه زنبورعسل به دلیل پراکندگی گسترده و همچنین ایفای نقش مستقیم در گرده افشانی گونههای زراعی، باغی و جنگلی از اهمیت مطالعاتی زیادی برخوردار است. به همین دلیل در این مطالعه چهار روش استخراج DNA شامل بهینه نمکی، فنل-کلروفرم، CTAB و CTAB+SDS به منظور گزینش مناسب ترین روش استخراج DNA زنبورعسل کوچک مورد ارزیابی و آزمایش قرار گرفت. سپس کیفیت و کمیت DNA استخراجی با استفاده از روش های اسپکتوفتومتری و ژل آگارز مورد مقایسه قرار گرفت. با توجه به کمیت و کیفیت DNA استخراج شده، روش CTAB+SDS برای استخراج DNA از زنبورعسل کوچک قابل توصیه است.

مناسب بودن گردههای گیاهی مختلف به تنهایی و در ترکیب با تخم کنه تارتن دولکهای (Tetranychus urticae Koch) برای کنه شکارگر Neoseiulus barkeri Hughes (Acari: Phytoseiidae)
در این پژوهش تاثیر رژیم های غذایی شامل گرده های ذرت، خرما، آفتابگردان، گردو، گرده زنبور عسل و طعمه طبیعی (کنه تارتن دولکه ای) در شرایط آزمایشگاهی (دمای 1±27 درجه سلسیوس، دوره روشنایی 16 ساعت روشنایی و 8 ساعت تاریکی و رطوبت 80-70 درصد) بر خصوصیات زیستی کنه شکارگر Neoseiulus barkeri Hughes مورد مقایسه قرار گرفت. درصد تلفات مراحل لاروی و پورگی کنه شکارگر در صورت تغذیه انحصاری از گرده های زنبور عسل و آفتابگردان صد در صد بود. طول دوره رشدی مراحل مختلف زیستی کنه شکارگر در صورت تغذیه با گرده های گیاهی مختلف با یکدیگر اختلاف معنی داری داشت. نسبت جنسی (نر+ماده/ماده) با تغذیه از گرده خرما (58٪)، کنه تارتن (77٪) و گرده های مورد آزمون+ کنه تارتن بیشتر بود. بیشترین نرخ ذاتی افزایش جمعیت در صورت تغذیه از گرده ذرت + کنه تارتن (244/0 بر روز) و گرده خرما + کنه تارتن (225/ 0 بر روز) بود. طول عمر افراد ماده با تغذیه از گرده خرما + کنه تارتن (12/12 روز) و گرده گردو (15/11 روز) طولانی تر از سایر تیمارها بود. طول عمر نر و ماده در صورت تغذیه از گرده طولانی تر بود و میزان تخمگذاری هم در صورت تغذیه از گرده + کنه تارتن بیشتر بود. کنه شکارگر در صورت تغذیه انحصاری از گرده ذرت، گردو و خرما مراحل زیستی را تکمیل کرده و قادر به تخمگذاری بود. بر اساس یافته های این تحقیق، گرده ذرت، خرما و گردو برای پرورش انبوه این کنه شکارگر مناسب بود و کنه شکارگر در صورت تغذیه انحصاری از گرده های ذرت، گرده گردو و گرده خرما قادر به زیست بود.
